Intended Use
For the quantitative in vitro determination of Glutathione Peroxidase in whole blood. This product is suitable for both manual and automated use.
General Background
Selenium is an essential trace element, involved in the aetiology of several diseases. At normal concentrations, selenium may have a protective effect against several diseases, including cancer. However, this protection is lost at low concentrations and selenium is highly toxic at high concentrations. Therefore, it is important that selenium levels should be monitored, so that they may be kept within the normal range. The Randox Ransel kit measures glutathione peroxidase which has a direct correlation with selenium levels.
Most clinical laboratories have established methods for determining GPX. However, because of differences in the concentrations of the various constituents used, it has been difficult to compare GPX results from one laboratory to another.
Principle
This method is based on that of Paglia and Valentine. Glutathione Peroxidase (GPX) catalyses the oxidation of Glutathione (GSH) by Cumene Hydroperoxide. In the presence of Glutathione Reductase (GR) and NADPH the oxidised Glutathione (GSSG) is immediately converted to the reduced form with a concomitant oxidation of NADPH to NADP+. The decrease in absorbance at 340 nm is measured.
