Clinical Significance
Free radicals are defined as chemical species that possess one or more unpaired electrons. Free radical attack has been linked to numerous pathological conditions in all organs of the body, such as cancer, diabetes, infertility, cardiovascular disease, lung, liver and renal disease, inflammatory and infectious conditions, ageing and neurological disorders. Antioxidants such as Superoxide Dismutase may account for an individual's ability to fight such disease.
Ransod allows the analysis of erythrocytic levels of SOD (Superoxide Dismutase) and may therefore be useful in the diagnosis of diseases associated with abnormal SOD levels, as a research tool in the assessment of new therapies and in determining the therapeutic efficacy and antioxidant potential of drugs.
Principle
The role of superoxide dismutase (SOD) is to accelerate the dismutation of the toxic superoxide radical (02), produced during oxidative energy processes, to hydrogen peroxide and molecular oxygen. This method employs xanthine and xanthine oxidase (XOD) to generate superoxide radicals which react with 2-(4-iodophenyl)-3-(4-nitrophenol)-5-phenyltetrazolium chloride (I.N.T.) to form a red formazan dye. The superoxide dismutase activity is then measured by the degree of inhibition of this reaction. One unit of SOD is that which causes a 50% inhibition of the rate of reduction of INT under the conditions of the assay.
