Intended Use
Immunoturbidimetric assay for the quantitative in vitro determination of alpha-1-antitrypsin in human serum and plasma. This product is suitable for use on a wide range of automated analysers.
Clinical Significance
Alpha-1-Antitrypsin is a glycoprotein made in the liver and plays a crucial role in protecting the lungs from damage caused by the enzyme neutrophil elastase. Emphysema is often the end result of such lung damage.
Alpha-1-Antitrypsin can therefore be used as a diagnostic marker to help diagnose emphysema especially in non smokers and individuals who have not been exposed to any lung irritants such as dust etc. The Alpha-1-Antitrypsin test can also used to determine a root cause for long term or persistent jaundice.
Alpha-1-Antitrpsin deficiency can be inherited with low levels in the blood (<80 mg/dl) indicating emphysema. Increased levels (3-4 times normal level) can occur as a result of trauma, pregnancy, administration of oestrogens or typhoid vaccine.
Principle
Patient samples are diluted and reacted with a buffer containing antibody specific for Alpha-1-Antitrypsin. The formation of the antibody-antigen complex during the reaction results in an increase in turbidity, the extent of which is measured as the amount of light absorbed at 340 nm. By constructing a standard curve from the absorbance of standards the Alpha-1-Antitrypsin concentration of a sample can be determined.
