Intended Use
For the quantitative in vitro determination of Creatine Kinase in serum or plasma. This product is suitable for use on the Hitachi 704, 705, 717, 902 and 911/912.
Clinical Significance
Creatine Kinase (CK) is a dimeric enzyme that catalyses the conversion of creatine to creatine phosphate, it is composed of two sub-units Muscle (M) and Brain (B) which associate to form three isoenzymes CK-MM, CK-MB and CK-BB. CK is present in many organs throughout the body but is perhaps found primarily in striated muscle, brain and heart tissues. Damage or disease to any of these tissues will result in elevated levels of the enzyme in the blood.
Determination of CK activity in both plasma and serum can therefore provide a sensitive and specific marker for myocardial infarction, hyperthyroidism, kidney disease, cerebrovascular accidents and skeletal muscle diseases such as muscular dystrophy.
The determination of CK using creatine phosphate and adenosine-5'-diphosphate (ADP) as substrates rather than creatine and adenosine-5'-triphosphate (ATP) has several advantages in test performance as it allows for a faster reaction rate resulting in greater sensitivity.
