Intended Use

For the quantitative in vitro determination of Leucine Arylamidase in serum, plasma and urine. This product is suitable for both manual and automated use.


Clinical Significance

Leucine Arylamidase (LAP) or Leucine aminopeptidase is a proteolytic enzyme that catalyses the hydrolysis of peptides containing leucine.

LAP is predominantly found in the cells of the liver however if the liver cells are damaged in anyway due to infection or drugs LAP is released into the bloodstream. Elevated levels of LAP may indicate jaundice, liver cirrhosis, carcinoma, and pregnancy. Unlike other liver enzymes LAP can also be measured in urine making it more convenient than other diagnostic markers.


Principle

Leucine arylamidase catalyzes the hydrolysis of L-leucine-p-nitroanilide to L-leucine and p-nitroaniline. The rate of formation of p-nitroaniline is determined by the rate of increase in absorbance at 405 nm.
