Background Information
The Beta Agonists (Beta-Adrenergic agonists) are a class of drugs commonly used to treat asthma however that have also been found to increase muscle growth and decrease fat deposition. Due to the nature and growth promoting properties of this group of drugs they are often used illegally by livestock producers in particular to increase the lean meat to fat ratio and improve feed conversion efficiency.
Inappropriate or illegal use of beta agonists in cattle and other food producing animals poses a serious risk to human health. Residues of the drug can be left behind resulting in contamination of meat as well as other food products from animal origin.
Principle
Sample and β-agonist labelled peroxidase conjugate are added to microtitre wells pre coated with purified antibody specific to β-agonist.
Any β-agonist in the sample will bind to the wells coated with antibody specific to β -agonist and consequently (antigen), (antibody) and (antigen peroxidase) complexes will be formed. After washing, One Shot Substrate is added to form a coloured complex. The intensity of the colour that may consequently develop is inversely proportional to the quantity of β-agonist present in the sample. The reaction is then stopped by the addition of Stop Solution and the resulting colour intensity can be read spectrophotometrically at 450 nm (ref 620 nm).
