Intended Use
For the quantitative in vitro determination of Boldenone in urine and serum. ELISA suitable for manual use.
Clinical Significance
Boldenone is a synthetic anabolic steroid traditionally used to treat debilitated, malnourished and underweight horses. It is derived from the naturally occurring hormone testosterone and it exerts a similar effect as testosterone and is very similar in structure.
In more recent time’s boldenone has been used illegally to improve the performance of racing animals or as a growth promoter to increase the lean meat to fat ratio in food producing animals. Due to the very long half life Boldenone drug residues can be detected in the blood up to a couple of months after use.
Principle
A microtitre plate is supplied pre-coated with a Boldenone antibody. Boldenone (antigen), if present in a standard or sample competes with horseradish peroxidase labelled Boldenone (enzyme labelled antigen) for a limited number of antibody sites on the microtitre plate. After incubation at room temperature to allow a competition reaction to take place, the microtitre plate is then washed to remove excess reagents. The enzyme substrate is added. After an incubation period to allow maximum colour development, the colour reaction is stopped by addition of Stop Solution. This produces a colour change from blue to yellow, and the absorbances are read at 450 nm. A standard curve is then constructed to determine the Boldenone concentration in the urine or serum sample.
