Intended Use

For the quantitative in vitro determination of Chloramphenicol in milk, milk powder, egg, honey, feed, tissue (including fish, fatty fish and prawn), serum and urine samples.


Clinical Significance

Chloramphenicol is a broad spectrum antibiotic used by the veterinary industry to treat bacterial infections. Chloramphenicol has been used extensively in apiculture to treat Bees with bacterial brood diseases such as American Foulbrood it has also been used in cattle to treat bacterial/viral pneumonias and pasteurella infections.

For bacterial and viral pneumonias as well as Epidemiological studies have raised concerns over the toxicity of Chloramphenicol associating its use with a number of serious diseases including aplastic anaemia, grey syndrome and bone marrow depression. Such toxicological problems have highlighted the importance of screening honey, meat and other animal products intended for human consumption for the presence of Chloramphenicol residues and metabolites. Currently the presence of Chloramphenicol in food producing animals within the EU is not tolerated.


Principle

Sample and chloramphenicol labelled peroxidase conjugate are added to microtitre wells pre-coated with purified antibody specific to chloramphenicol.

Any chloramphenicol in the sample will bind to the wells coated with antibody specific to chloramphenicol, consequently antigen/antibody and antigen/peroxidase complexes will be formed. After washing, One Shot Substrate is added to form a coloured complex. The intensity of the colour that may consequently develop is inversely proportional to the amount of chloramphenicol present in the sample. The reaction is then stopped by the addition of Stop Solution and the resulting colour intensity can be read spectrophotometrically at 450nm (ref 630nm).
