Clinical Significance

Quinolones are synthetic antibiotics that inhibit the activity of DNA gyrase and topoisomerase IV. The fluoroquinolones are active against enterobacteriaceae, also staphylococci, P. aeruginosa, Mycoplasma, Chlamydia and some streptococci.1 Quinolones are widely distributed to most body fluids and tissues. They are variably metabolised in the liver and excreted in the urine. The presence of Fluoroquinolones in foods has become of increasing concern as studies indicate the presence of antibiotic resistant pathogens2. Multiple countries are considering bans on their use. Honey has been found to be adulterated with antibiotics, which has led to import bans2. Fluoroquinolones are not recognised as safe or effective to treat honey bee diseases and they have not been approved for such use3.

This ELISA aims to give a fast, reliable, analytical method to detect the presence of flumequine. If performing the test using duplicate wells, this kit has the ability to analyse 40 samples.


Principle

A microtitre plate is supplied precoated with flumequine antibody. Flumequine (antigen), if present in the sample competes with the horseradish peroxidase labelled flumequine (enzyme labelled antigen) for a limited number of antibody sites on the microtitre plate. After incubation at room temperature to allow a competition reaction to take place, the microtitre plate is washed to remove excess reagents.

The enzyme substrate is added. After an incubation period to allow maximum colour development, the colour reaction is stopped by the addition of acid. This produces a colour change from blue to yellow, and the absorbances are read at 450nm.

A standard curve is then constructed to determine the flumequine concentration in the sample.
