Intended Use
For the quantitative in vitro determination of phenylbutazone and oxyphenbutazone in serum and plasma samples.
General Background
Phenylbutazone is a pyrazolone derivative and one of the first non-steroidal anti-inflammatory drugs (NSAIDs) to be approved for veterinary use in horses and dogs.
The use of phenylbutazone in food producing animals is prohibited in most countries, including the European Union, United States, Canada and Japan. As a result, MRLs have not been set for phenylbutazone residues, meaning that any detection is considered a violation.
Principle
Sample and phenylbutazone labelled peroxidase conjugate are added to microtitre wells precoated with purified phenylbutazone antibody
Any phenylbutazone/ oxyphenbutazone in the sample will compete with the phenylbutazone labelled conjugate for binding sites on the phenylbutazone antibody.
After washing, One Shot Substrate is added to form a coloured complex. The intensity of the colour that may consequently develop is inversely proportional to the quantity of phenylbutazone/ oxyphenbutazone present in the sample. The reaction is then stopped by the addition of Stop Solution and the resulting colour intensity can be read spectrophotometrically at 450 nm (ref 630 nm).
