For the quantitative in vitro determination of Streptomycin and Dihydrostreptomycin in honey. For manual use only.


General Background

Streptomycin and dihydrostreptomycin are aminoglycosides, antibiotics which act by binding to the 30S ribosome and inhibit protein synthesis. They are used mainly to treat infections caused by aerobic gram-negative bacteria although they also act synergistically against some gram-positive bacteria. Aminoglycosides have also been used as growth promoters in food producing animals and there has been controversy surrounding this due to the potential for the development of aminoglycoside resistant pathogens. Traces of Streptomycin have been found in honey samples worldwide.

This ELISA aims to give a fast, reliable, analytical method to detect the presence of Streptomycin and Dihydrostreptomycin. This kit can be used as a screening method and will give results in 1½ hours (not including sample and reagent preparation). If performing the test using duplicate wells, this kit has the ability to analyse 40 samples.


Principle


A microtitre plate is supplied precoated with Streptomycin antibody. Streptomycin (antigen), if present in the standard and sample compete with horseradish peroxidase labelled Streptomycin (enzyme labelled antigen) for a limited number of antibody sites on the microtitre plate. After incubation at room temperature to allow a competition reaction to take place, the microtitre plate is washed to remove excess reagents.

The enzyme substrate is added. After an incubation period to allow maximum colour development, the colour reaction is stopped by addition of acid. This produces a colour change from blue to yellow, and the absorbances are read at 450 nm (ref 630nm).

A standard curve is then constructed to determine the Streptomycin concentration in the standard and sample.
