Intended Use

For the quantitative in vitro determination of Alkaline Phosphatase (ALP) in serum or plasma. This product is suitable on the Hitachi 717, 911 and 912 analysers.

Clinical Significance

Serum Alkaline Phosphatase measurements are of diagnostic use in the investigation and treatment of hepatobiliary disease and bone disease associated with increased osteoblastic activity. Also useful in diagnosis of parathyroid and intestinal diseases.

Colorimetric Method

This is an optimised standard method according to the recommendations of AACC and IFCC.

Principle

The substrate p-nitrophenyl phosphate is hydrolyzed by Alkaline Phosphatase from the sample, in the presence of Magnesium ions, to form p-nitrophenol which is yellow in colour and can be read at 405 nm.

The intensity of colour produced is proportional to the Alkaline Phosphatase activity in the sample.
