Intended Use

For the quantitative in vitro determination of Acetaminophen in human serum. This product is suitable for use on the RX series instruments which includes the RX daytona and RX imola.

Clinical Significance

Acetaminophen is a commonly used analgesic drug, which usually has no adverse effects. However it has been found in some cases of long-term treatment with acetaminophen, hepatotoxicity and nephrotoxicity have been reported where chronic excessive use has been a factor. This assay is also used in the detection of acute overdose which can cause severe hepatic damage leading to hepatic failure if left untreated.

Principle

The enzyme, arylacylamidase contained in R1, hydrolyses acetaminophen by cleaving the amide bonds of the acetaminophen molecule to produce p-aminophenol and acetate. The p-aminophenol is subsequently converted to an indophenol in the presence of o-cresol, where periodate contained in R2 is used as a catalyst, and the reaction is measured colorimetrically at 600nm.
