Intended Use

For the quantitative in vitro determination of Transferrin in Serum. This product is suitable for use on the Hitachi 704, 717, 902 and 911/912.


Clinical Significance


Transferrin (siderophilin) is the principal iron binding protein in human plasma and can bind two molecules of iron; it is responsible for transporting iron to the liver, spleen and bone marrow. Plasma levels of Transferrin are regulated by the availability of iron and increase when plasma iron is low. Levels correlate well with the Total Iron Binding Capacity (TIBC) of serum.

Decreased levels of transferrin in the blood may be hereditary (atransferrinemia) or due to poor production of transferrin in the liver or a loss of transferrin through the kidneys. Transferrin is extremely important in the production of hemoglobin which is impaired when transferrin levels are low, low levels can therefore lead to anemia in some individuals.

The normal range for healthy adults is between 200-400 mg/dl with levels also increased during pregnancy, estrogen administration, iron deficiency, inflammation, malignancy, liver disease, malnutrition and protein loss.


Principle

Sample is reacted with a buffer containing antibody specific for human transferrin (siderophilin). The absorbance (600 nm) of the resulting turbid solution is proportional to the concentration of transferrin in the sample. By constructing a standard curve from the absorbance of standards, transferrin concentration of sample can be determined.
